Kisuke initially serves as Karakura Town’s source of spiritual equipment and information. He sells Rukia replacement supplies, gives Ichigo access to his Soul Reaper form and explains supernatural events without immediately revealing his own history. When Byakuya Kuchiki removes Ichigo’s borrowed power, Kisuke offers to restore it through a dangerous training process beneath the shop.

The training nearly transforms Ichigo into a Hollow, but it also awakens his own Soul Reaper power and leads him to learn Zangetsu’s name. Kisuke then supplies the Senkaimon required for Ichigo, Orihime, Chad, Uryū and Yoruichi to enter Soul Society. He remains in the Human World during the rescue, partly because a barrier established after his exile prevents him from returning by ordinary means.

The Soul Society conflict reveals that Kisuke hid the Hōgyoku inside Rukia’s soul. Aizen engineered Rukia’s execution to extract it, making Kisuke’s attempt at concealment one of the conditions behind the crisis. Once Aizen obtains the Hōgyoku and defects, Kisuke’s warnings and preparations become central to the conflict that follows.

During the Arrancar campaign, Kisuke defends Karakura Town, studies Aizen’s evolving forces and helps train Ichigo’s allies. He confronts Yammy Llargo, protects the Human World from additional incursions and opens the Garganta used to reach Hueco Mundo. He later joins Yoruichi and Isshin against Aizen, combining Kidō, physical attacks and specialized devices in an attempt to stop his transformations.

Kisuke’s attacks do not overpower Aizen directly. Instead, he implants a seal that activates after Ichigo’s Final Getsuga Tenshō weakens Aizen and the Hōgyoku begins rejecting him. The seal ends the battle and permits Soul Society to imprison Aizen, completing the countermeasure Kisuke had prepared around the possibility that raw force would be insufficient.

In the Thousand-Year Blood War, Kisuke investigates the Wandenreich’s methods and coordinates operations across Soul Society, the Human World and Hueco Mundo. He determines that Quincy cannot steal Hollow-tainted Bankai and distributes Shin’eiyaku pills that temporarily Hollowfy the captains’ Bankai, allowing them to reclaim their weapons. He later creates a gateway into the transformed Royal Palace and leads part of the assault on Wahrwelt, where his battle with Askin finally reveals Benihime’s Bankai.
