Mako is cheerful, affectionate, energetic, distractible, and almost impossible to intimidate for long. Her thoughts frequently leap between unrelated subjects, producing elaborate speeches accompanied by equally elaborate gestures whenever she believes Ryuuko needs encouragement.
She possesses an uncomplicated capacity for accepting people. Mako decides almost immediately that Ryuuko is her friend and rarely questions that conclusion regardless of how dangerous Ryuuko's battles become. Her loyalty is particularly important because she approaches Ryuuko as an ordinary friend rather than as a fighter, Life Fiber user, or political symbol.
Mako is not completely immune to temptation. When Honnouji's hierarchy gives her family an opportunity to escape poverty, she discovers how quickly wealth and status can change their priorities. Her eventual rejection of those privileges demonstrates that friendship matters more to her than the social rank she once enthusiastically embraced.

Mako is normally an ordinary student without the enhanced abilities provided by a Goku Uniform. Her unusual durability and physical feats are generally presented through the series' exaggerated comedy rather than as a formally defined supernatural power.
Her most effective ability is her capacity to reach Ryuuko emotionally. Mako's intervention can interrupt Ryuuko's rage or self-destructive behavior when physical force fails, and her seemingly nonsensical speeches often identify exactly what Ryuuko needs to hear.
Mako temporarily receives a Two-Star Goku Uniform as president of the Fight Club. The delinquent-styled uniform grants her dramatically enhanced strength, speed, and durability, allowing her to fight at a level far beyond her ordinary capabilities. She later receives another version of this uniform for the battle aboard the Naked Sol.

Mako befriends Ryuuko on her first day at Honnouji and quickly becomes involved in her confrontations with the student council. Her family invites Ryuuko to live with them, making Mako the central connection between Ryuuko's violent investigation and ordinary daily life.
During the Fight Club episode, Mako becomes club president while Ryuuko wins matches on the club's behalf. Their success rapidly elevates the Mankanshoku family's social status. Mako initially becomes consumed by the resulting wealth and privilege but ultimately disbands the club rather than sacrifice her friendship with Ryuuko.
Mako continues accompanying Ryuuko throughout the conflict surrounding Honnouji, repeatedly intervening when her friend becomes isolated or overwhelmed.
When Ryuuko learns the truth about her family and later falls under Junketsu's control, Mako remains one of the people most determined to bring her back. She eventually fights alongside the combined forces opposing Ragyou and supports Ryuuko through the final Life Fiber crisis.

Ryuuko Matoi is Mako's best friend, a designation Mako applies almost immediately and never seriously withdraws. Her unwavering acceptance repeatedly helps Ryuuko Matoi recover when anger or despair isolates her from everyone else.
Barazou Mankanshoku and Sukuyo Mankanshoku are Mako's parents. Their chaotic household becomes Ryuuko's home after she arrives in Honnou City.
Matarou Mankanshoku is Mako's younger brother. The siblings share their family's energetic temperament despite frequently bickering.
Guts is the Mankanshoku family's dog and constant participant in the household's obsession with food.
Ira Gamagoori begins as an intimidating authority figure at Honnouji but repeatedly becomes involved in protecting Mako. The series strongly suggests that Gamagoori develops romantic feelings for her, although their relationship is never formally established as a couple.
Satsuki Kiryuuin initially represents the school hierarchy threatening Ryuuko and Mako. After Satsuki joins Ryuuko against Ragyou, Mako accepts her as an ally with characteristic ease.
