Tatsumi commands the Yaminobu operation targeting Battōsai. His organization obtains intelligence from within the Chōshū faction and develops a plan in which Tomoe Yukishiro becomes central to discovering and exploiting Kenshin's weakness.

Tatsumi deliberately allows Kenshin and Tomoe's emotional attachment to deepen, reasoning that Tomoe herself can become the weakness needed to destabilize Battōsai. When Kenshin enters the Yaminobu forest to find her, Tatsumi's men progressively damage Kenshin's senses before the final confrontation.

Tatsumi ultimately fights the badly impaired Kenshin himself. Tomoe intervenes during the decisive exchange, and Kenshin's strike cuts through both her and Tatsumi, killing them.

Tatsumi's influence extends into the Jinchū Arc through his former student Banjin Inui, who joins Enishi's campaign partly to avenge his master's death.
