Nezuko's transformation creates the central objective of the series before Tanjirou ever joins the Demon Slayer Corps. Rather than beginning as a conventional mission to avenge murdered relatives, the story gives him a living family member whose condition may still be reversible. VIZ's official synopsis explicitly frames his journey around returning Nezuko to normal.

For much of the opening storyline, Nezuko travels inside the box on Tanjirou's back during daylight. When demons attack, she frequently becomes his closest combat partner. Her ability to fight without consuming humans provides continuing evidence that demon behavior is not completely immutable.

The Asakusa storyline expands the possibilities surrounding her condition when Tanjirou meets Tamayo. A demon capable of medical research gives him something he did not possess during his training: an actual scientific route toward developing a cure.

Mount Natagumo brings Nezuko into one of the anime's most important early battles. When Rui threatens Tanjirou, she awakens Exploding Blood and combines it with her brother's Hinokami Kagura. The battle also brings her existence to the attention of the Hashira and forces the Corps to decide whether she should be treated as an exception.

The Hashira meeting places Nezuko's self-control under direct scrutiny. Sanemi deliberately tempts her with his unusually attractive blood after injuring her. Her refusal to attack him becomes one of the strongest demonstrations that her relationship with humans differs fundamentally from that of ordinary demons.

During the Entertainment District battle, Nezuko becomes a major combatant in her own right. Her mature demonic form allows her to overwhelm Daki temporarily, while Exploding Blood proves capable of neutralizing Gyutaro's demon-generated poison.

The Swordsmith Village battle changes her importance to the wider demon conflict. Tanjirou is forced into a choice between pursuing the fleeing Upper Rank demon and protecting Nezuko as sunrise approaches. Nezuko physically forces him toward the enemy, choosing the lives of endangered villagers over her own apparent survival.

Instead of disintegrating, she remains beneath the sun. Her newfound immunity establishes a phenomenon neither the Demon Slayer Corps nor Muzan believed possible and radically changes the strategic importance of the Kamado siblings.

After becoming capable of surviving sunlight, Nezuko becomes Muzan's desired target because consuming her may allow him to acquire the same immunity. The Corps therefore has strong reason to keep her away from the Infinity Castle battle.

Tamayo's medicine eventually restores Nezuko's humanity while Tanjirou and the Demon Slayers are engaged in the final battle. She travels toward him as her memories and ability to speak return.

The siblings' roles briefly reverse after Muzan transforms Tanjirou into a demon. Nezuko, now human, refuses to abandon her brother despite his attacks and becomes one of the people trying to bring him back rather than kill him.
