Kohaku was born in Ishigami Village in 5723, thousands of years after the petrification of modern humanity. She is the younger daughter of village chief Kokuyo and the younger sister of priestess Ruri. The village's isolation gives her no access to modern education or medicine, but it develops her tracking, gathering and wilderness skills from childhood.

Ruri's chronic illness defines Kohaku's early life. Rejecting the expectation that her sister will simply die young, she repeatedly carries heavy jars of hot-spring water across difficult mountain terrain to ease the symptoms. The routine strengthens her body and demonstrates the persistence she later recognizes in scientific work.

Kohaku first observes Senku Ishigami during his confrontation with Tsukasa Shishio. Believing Senku has been killed, she attacks Tsukasa and is defeated, then trapped beneath a fallen tree. Senku returns and constructs a pulley system to free her, choosing a laborious scientific solution over leaving her behind.

Impressed by his courage and inventions, Kohaku brings Senku toward Ishigami Village. She becomes his first firm ally among its people and introduces him to the local social order, craftspeople and resources. Her determination to cure Ruri supplies the Kingdom of Science with its first large shared objective: producing an antibiotic from Stone World materials.

Kohaku is direct, energetic and impatient with passivity. She responds quickly to danger and prefers concrete action to prolonged debate. Her protective instinct is strongest around vulnerable people, especially Ruri and Suika, but it gradually expands to include the entire Kingdom of Science.

She wears formal clothing and cosmetics for an infiltration on Treasure Island, insulated gear during global travel and a spacesuit for the lunar expedition. Kohaku is one of Ishigami Village's strongest fighters and its fastest prominent warrior. Supplementary statistics identify speed as her best combat attribute, supported by agility, balance, reflexes and stamina. She can cross difficult terrain quickly, climb, swim, carry heavy loads and react to attacks before less experienced fighters register them.

Her eyesight is extraordinary even by the heightened standards of the series. Kohaku identifies distant people and objects, detects small movements and serves as a lookout during exploration. Senku Ishigami earns Kohaku's trust by saving her with a pulley and committing himself to Ruri's cure. Their abilities remain complementary: Senku Ishigami supplies scientific roadmaps, while Kohaku supplies labor, senses, field judgment and protection. She readily identifies the kindness behind his utilitarian explanations. Their interactions contain teasing and romantic ambiguity, but the main manga does not establish them as a couple.

Suika is treated by Kohaku as a younger sister and capable teammate rather than a burden. Kohaku protects Suika during the village tournament and encourages her participation in Kingdom projects. Chrome is a childhood companion whose “gorilla” remarks provoke arguments; beneath the bickering, Kohaku respects his devotion to science and Ruri. Tsukasa Shishio first appears to Kohaku as an overwhelmingly powerful enemy, but they later fight together with mutual professional respect. Hyoga also moves from dangerous opponent to conditional ally, and Kohaku never confuses cooperation with complete personal trust. Stanley Snyder's firearms place him outside the kind of contest her village training can answer directly; after the continental conflict, the marksman's specialized abilities make him her fellow crew member on humanity's lunar mission.

Kohaku is the first person born in the Stone World to commit fully to Senku's project. Through her, he gains access to Ishigami Village, while the reader sees how a person without modern schooling responds to reproducible science. Her physical rescue by pulley power establishes the division of labor that defines their alliance.

The effort to cure Ruri makes Kohaku's private hope the emotional center of the Kingdom of Science's first industrial roadmap. She gathers materials, protects workers and helps recruit villagers while Senku and Chrome build the chemical chain. During the campaigns in North and South America, firearms reduce the tactical value of open melee combat, so Kohaku works through stealth and coordinated assault. She is mortally shot during Stanley's final attack before a worldwide petrification wave preserves the battlefield; Suika later recreates revival fluid and restores the group. Kohaku subsequently joins Senku and Stanley on the lunar expedition, carrying the perspective and physical skills of Ishigami Village beyond Earth.

Kohaku begins as a solitary protector who expects to carry impossible burdens herself. Her repeated hot-spring journeys for Ruri are admirable but cannot cure the underlying disease. Senku's arrival does not replace her effort; it places that effort inside a system where knowledge, craft and cooperation can produce a result strength alone cannot.

The Kingdom of Science teaches her to trust specialization. She learns that Chrome's experiments, Kaseki's hands, Gen's persuasion and Suika's unobtrusive scouting can matter as much as combat. Her later selection for the Moon mission completes the expansion of her world. A teenager defined by one sister and one isolated village becomes a representative of humanity in space. The change does not diminish her attachment to Ruri or her instinct to protect others; it gives those traits a global scale and a more disciplined method.

Kohaku is the Japanese word normally written 琥珀, meaning “amber.” Her name is written phonetically in katakana as コハク in the series. The mineral meaning fits Ishigami Village's broader use of names associated with stones, minerals and materials.

Amber is fossilized tree resin rather than a mineral in the strict geological sense. That distinction suits a naming system built around familiar material terms rather than formal mineral classification. No separate family name is used for Kohaku in the main story.

The nicknames “lioness” and “gorilla” refer to her ferocity and physical strength. They are informal descriptions used by other characters, not titles she selects for herself or names of combat forms. English wording varies with translation and adaptation.

Kohaku's eyesight is given as 11.0 in official fanbook data, far beyond ordinary human visual acuity. The ability makes her a natural lookout and allows her to distinguish details that other characters require lenses to see.

Her favorite food is listed as meat dishes, while her personal treasure is the hair ornament from Ruri. The official anime site celebrates her birthday on August 8.

Manami Numakura voices her in Japanese and Felecia Angelle in English. The final season completes the original manga's principal story.

The anime adds performance, timing and transitional action but does not give Kohaku a separate biography or formal set of named techniques.
